Hasan Johnson 02. 23. 13 Extra Credit The Movie â€Å"Django Unchained† The objective of this paper is to review some of the results of the discussion-panel’s analysis of certain aspects of this movie that relate to the Hip-Hop culture. On Thursday, February 21, 2013, I attended a panel discussion hosted by the Africana Studies Department of Fresno State University. It was conducted at the African American Museum, Fresno California. Its purpose was to evaluate this movie’s effect on us (basically Black-Americans), by analyzing elements of this movie.This movie presents Black masculinity and femininity during the 1860s slavery era of America using characters that portray slaves as childish and stupid (the Betina or D'Artagnan), violent (the Mandingo or Django), hypersexual (Sheba) and cowardly (all the black male slaves). These slavery created representations of Black masculinity (the Buck) and femininity (the Jezebel/Mulattoe) are relevant to Hip-Hop because they are the predominate characters being proffered as entertainment in the overwhelming majority of Hip-Hop videos.The stereotypical persona of â€Å"The Buck† in the videos is the Gangsta/Thug and â€Å"The Jezebel/Mulattoe† is the Bitch/Ho— both are slavery created stereotypes. The Jezebel/Mulatto (Bitch/Ho) The Buck (Gangsta/Thug) Another aspect of this movie relevant to Hip-hop culture is the word â€Å"nigger†, used incessantly in the movie, just as, its morphed version, â€Å"Nigga† is used incessantly by Hip-Hoppers, in conversation and in their videos.Since language is one of the seven essential element of all cultures, the usage of the word â€Å"nigger† was discussed by the panel and audience and as a result of this discussion, we agreed that—without the use of the word â€Å"nigger†, â€Å"Django Unchained† would not be authentic; because, in this movie, â€Å"nigger† is a descriptive term, used to describe property, e. g. a slave or slaves—with no malice intended. Here are two examples, horse or horses and house or houses—ergo, nigger or niggers.The only character that uses â€Å"nigger† other than as a description of property is Stephen (the â€Å"Uncle Tom† played by Samuel L. Jackson). He used the word â€Å"nigger† to degrade his fellow black slaves. As with the movies use of â€Å"nigger†, the same can be said about Hip-Hop videos, they would lose its authenticity without the use of the word â€Å"Nigga† because â€Å"Nigga† in Hip-hop is a descriptive turn, but unlike â€Å"nigger† it versatility. For example, it can be used describe a relationship and its meaning changes depending on who is saying it and to whom.For example, in the movie when a Slave-master says â€Å"My nigger†, it does not have the same meaning as a Gangsta/thug or Jezebel/Mulatto e in a Hip-Hop video saying, â€Å"My Nigga†. The Slave-masters are describing their property, while the Hip-Hoppers are describing their relationships. During the audience participation portion of this discussion, a young black- man in the audience (Christopher Jackson) described how much the movie upset him.He also shared with us, the extent to which it made him realize how horrible slavery was; and how much suffering his ancestors had to endure; so that he can enjoy the civil rights he has today. A glance at the statistics stated in the policy at the time of its reading, show that the mechanisms that the federal government intended to adopt were the most satisfying. In general, teachers explain the content of textbooks, hand-outs, and other materials. But in American schools, teachers think class participation is very important, and they require students to give their opinions or ask questions in classes. Another difference is that teachers in Japan write down important things on a blackboard, but teachers in the U.S. seldom write everything; they just explain orally. Japanese students are very quiet during classes, and all they do is listen to teachers and copy what teachers write on blackboards. Milton and Cavendish: Faithful Realists :: Paradise lost Blazing World

Milton and Cavendish: Faithful Realists Inquiries regarding the nature and acquisition of knowledge, coupled with the monumental question of whether human beings are capable of accruing knowledge–the philosophical study of epistemology–has roots buried in antiquity: Genesis, to be exact. Great thinkers of the Western tradition have both accepted and rejected components of Old Testament lore; Platonic and Aristotelian philosophers have indeed battled for centuries over the way in which reality is understood. Following Aristotle’s teachings, the empiricists and Enlightenment thinkers regarded the processing of sense and experiential data as the surest way to unlock truth. Plato’s adherents, however, figures such as Immanuel Kant, deemed the human intellect a leaky and misguiding faculty, not quite efficient in comprehending truth. John Milton and Margaret Cavendish, the reigning theological epistemologists of the 17th century, pondered the nature of divine reality, the role of human rationality in understanding God’s master plan, and the means by which that plan is (and should be) grasped by the human race. Both Milton and Cavendish have declared in their works, Paradise Lost and The Blazing World, that reason as a means to arrive at ultimate truth is insufficient; in the end, faith is the only tool with which human beings acquire proper knowledge. After an initial reading of The Blazing World, one would assume Cavendish ranked reason above faith, parting ways with Milton; the Empress in the tale is nearly obsessed with scientific inquiry. Upon close analysis of the text, however, it becomes evident that Cavendish’s message is complementary to Milton’s. This is not to say that either Milton or Cavendish were pure theologians in their world view, placing no value on science or logic; rather, both found a measure of importance in the findings of contemporary science and consequently instilled in their literary protagonists curiosity about the laws of the universe. It was just such cosmic curiosity that plagued thinking individuals of the Renaissance period. As Europe slowly developed a flavor for scientific inquiry, well guarded theological dogmas were threatened; the mid 1600s was indeed a time of questioning long established religious and political doctrines. While grappling with the emerging debate of reason versus faith, Milton and Cavendish offered philosophical fictions heralding the supremacy of the latter. Characters in the authors’ works discover that reason, untempered by belief in divine truth, is dangerous. Cavendish’s Empress of the Blazing World, for example, is a tyrannical ruler who demands that her subjects uncover the secrets of the natural world.

A. E. Housman’s “To An Athlete Dying Young” Essay

E. Housman’s â€Å"To An Athlete Dying Young† is part of his series of poems collected in his book, A Shropshire Lad. It is set during the funeral march of a young and victorious runner who died at the height of his career. The speaker is one of the pallbearers, who recounts the athlete’s victory and philosophizes on the timeliness of his death. As with the rest of the book, this poem resounds with the melancholic themes of death and dying in youth. To be more specific, this poem describes how it is best to die young and at the peak of one’s career in order for one’s memory and legacy to be forever etched in the minds of the people. Though Housman’s exhortation might seem to be bordering on the macabre, it rings true: glory, fortune, and fame are all fleeting. The poem starts by depicting the victory of the runner and how he became the source of pride in his town: â€Å"The time you won your town the race  / We chaired you through the market-place.† The image of chairing shows how the townspeople carried the runner on their shoulders and paraded him in the market-place. The market setting is important, as it depicts a place where people of all ages and social status converge. It is a testament to the runner’s achievement and how it was able to bring together different people together in his victory. As they were carrying him on his shoulder, â€Å"Man and boy stood cheering by, / And home we brought [him] shoulder-high.† The last line of the first stanza foreshadows what would happen as the poem progressed. Carrying him shoulder-high not only indicates how he was chaired by the townspeople but also foreshadows how his coffin would be carried at the same shoulder-high level as he is brought to his tomb. Also, home could be read two ways given the foreshadowing: home as in his physical house and home as in his final resting place. In this manner, the speaker draws a connection between his victory and his death, and how, in both instances, he would be carried by the shoulders to be brought â€Å"home.† In the second stanza, the procession takes a sudden turn from one of gaiety and victory to one of sadness and melancholy, as this time, they are no longer celebrating but sending the athlete to his grave. â€Å"Shoulders† and â€Å"house† again are mentioned, paralleling that of the first stanza albeit taken on a somber tone. The procession takes place on the â€Å"the road all runners come,† a metaphor for the road of life that everyone travels. Another metaphor makes an appearance to further the sober theme: once the athlete has been brought â€Å"home,† he becomes a â€Å"townsman of a stiller town.† The â€Å"stiller town† here refers not only to the cemetery at which he is buried but to the idea that he is resting in peace, that he has accepted his death. Again, this idea of peaceful death corresponds well to the theme. When young people die, people always assume that it is â€Å"untimely,† hence, they cannot rest in peace. However, for the athlete, his death is timely as he is at the peak of his career, ensuring that he will remain a glorious image in the minds of the townspeople for generations to come. The third stanza presents the main theme of the poem by saying that the runner was a â€Å"smart lad, to slip betimes away / From fields where glory does not stay.† The speaker also presents here the laurel as a symbol for glory and the rose as a symbol for the life of non-victors. He compares that lifespan of the laurel to the rose, indicating that the laurel easily withers, while the rose, slow to bloom, lives much longer. The fourth and fifth stanza both expound on the third, as it gives evidence of how death is indeed a blessing for the young athlete, for in death, he is able to preserve his victory. In death, he â€Å"cannot see the record cut,† indicating that he will not have to deal with younger and better runners eventually overtaking him, as they will inevitably do. In dying, too, he will not become like any run-of-the-mill athlete, who did not quit while they were ahead. As such, he is a privileged position, as he will not live to be disgraced, for his body died before his name. In the third stanza, death is represented by the metaphor â€Å"shady night,† which works on an age-old technique of associating death with night. An oxymoron is also used in the third stanza – silence sounds. It marries together two contradictory terms, which furthers the image of death, for in death, the only â€Å"sound† to be heard is â€Å"silence.† The main point of these two stanzas is that the brief life that he had is esteemed, for he did not have to suffer the loss of fame and glory. In the fifth stanza, the athlete is about to be laid in his tomb, and the speaker is exhorting his companions to recount once more the victory. He rationalizes that in doing such an act the memory of the runner’s victory is immortalized, preserving it in the minds of the people. The symbol of the challenge-cup is also used as a testament to the glory of the athlete. Once again, the metaphor of the death as shade is used, indicating that death is not the loss of the light of life but merely the blocking of light. The importance of this metaphor to the poem is that death cannot extinguish the light of the runner’s glory. At most, it can only dim it to a more subtle shadow. Finally, the last stanza elevates the theme of the poem – as well as the glorious athlete – towards another, spiritual level. In a way, the last stanza encapsulates the promise of dying young and at the height of fame, and that despite the seeming â€Å"untimeliness† of his demise, much more is to be gained through death. As the athlete passes on to the next life, the â€Å"strengthless dead† will â€Å"flock to gaze† at his â€Å"early-laurelled head.† The image of the dead surrounding the young athlete (implied by the phrase early-laurelled) is a very powerful one, as it indicates how the glory of the athlete transcends the mortal plane, which makes it an even more laudable success. Furthermore, in this spiritual state, his laurel crowns will not wither. What this means is that his success has finally become immortal, and his glory will no longer fade, preserved as it is, ironically, through death. Death as a way to preserve glory and success may seem to be too vain and too gruesome. However, it is important to go beyond this theme and instead, reach beyond the heart of the poem. Spring Jazz Band Concert Report

On Friday, April 29, 2011, I went to see Spring Jazz Concert at Sophia B. Clarke Theater. Most of performers have great instrument skill and show us a wonderful music. Each movement highlights the character, range, and technical abilities of an individual instrument, all of pieces stand on their own as intricate musical gems. The show was divided for two parts, and they have different sections on those two parts.During the concert, we would hear different music programs by playing in different music instruments such as saxophones, trumpets, trombones, drums, bass, guitar, and piano. Each performer represents different music instruments. On the first parts of the concert, there were three songs. In conclusion, I enjoyed this concert very much because the music in this concert was very nice. In my opinion, I had totally different feelings between I listened music in the concert and at home, and the atmosphere of concert was awesome.Besides the music, I had deep impression in the perfor mers; every performers in the concert had much passion when they were performing. In my opinion, because of their wonderful performance, I could hear such nice music. Also, everyone in the concert was polite. Every times when conductor came to the stage or the song was end, almost every audience stood up and applauded to the performers, the applause lasted for a pretty long time. At that time, I felt so good. All in all, I really enjoyed this concert.The tone color of most of the jazz bands that I saw was smooth and inviting. This made the bands easier to listen to. They were polyphonic in structure, where all of the different instruments were playing different melodies. The rhythm that was used was in a quadruple meter. This was true for all of the bands. The only thing that I wished would have happened was that there would have been more improvisation instead of reading from sheet music. They were very structured in that way.

Farmers

Farmer’s discontent during the period 1870 – 1900 had an impact on their attitudes and actions towards national politics. During this time period manufacturing had a huge growth spurt and agricultural began to decline. This made it harder for farmers to make a living. Document G shows how much manufacturing increased between the years 1849 – 1899. America could no longer aspire to be a nation of small freehold farms. Manufacturers and people living in big cities depended on farmers to pretty much keep them alive. Many people didn’t realize how much of an affect farmers had on their everyday lives. If you took farms away from the United States during this period of time, everything would have completely crashed. Farmer’s had difficulties making a living because the rates of being a farmer were so high. â€Å"Nothing has done more to injure the western region than these freight rates. † (Document F) The high rates of being a farmer made it difficult to pay payments on the lands and the rate of interest was rapidly rising. Some farmers couldn’t even sell their produce at a reasonable profit. These farmers worked long, hard hours and the government wasn’t on their side about paying them with a decent income yet they wanted so much from the farmers. Along with farm prices failing, railroad prices were increasing. Railroads were important to farmers because they took farmers out to their lands, carried their produce of hard work to markets, and brought them the manufactured goods that they needed. Many farm settlements were established around railroads because of this reason. Railroad managers were forced to charge very high rates and because of it, that means farmers would have to pay more money to use railroads when they already used enough money keeping their crops alive. Document B talks about how it was also hard for colored farmers to make a living especially after the Civil War. â€Å"They had to get the local merchant or someone else to supply the food for the family to eat while the first crop was being made. (Document B) They couldn’t pay interest and by the end of the first year, they found themselves in debt. The attempt in the second year didn’t go so well either because they had old debts plus new interest to pay. â€Å"Many of the colored farmers had almost given up hope†¦Ã¢â‚¬  (Document B) Not only was money an issue for farmers but also in the late 1880s and early 1890s crop suffered severely from droughts. It was cool and dry which was unfavorable for all crops. This made it difficult for farmers to keep their crops alive. By the late 1880s, farmers were so fed up with everything that they created the Farmers Alliance, seeking to overthrow the chains from the banks and railroads that bound them. The Farmers Alliance merged with other liberal Democrats to form the Populist Party. They strived for railroad regulation, tax reform, and unlimited coinage of silver. In the election of 1892 the Populist Party nominated James B.  Weaver for president and as Document D shows, most of the farmers in the west, mainly Nevada, voted for the Populist Party because that party was rooted in the frustration of the farmers and they needed change. The farmers were furious with the way things were going for them and they had hoped that the Populist Party would help. Even though they didn’t win the election, they made a remarkable showing with 1,029,846 popular votes and 22 electoral votes. These electoral votes only came from the midwestern and western states. With the creation of a new party, out of discontent, the farmers managed to change national politics.

Classical studies Essay The Colosseum was the first permanent amphitheatre to be built in Rome. Its huge size, as well as the practical and efficient way it dealt with organising of events and ways of controlling large crowds in a safe manner makes it one of the greatest architectural buildings ever constructed in the Roman Empire and was a gift from Emperor Vespasian and finished under Emperor Titus. The building itself is a vast ellipse with tiers of seating for about 50,000 spectators around a central elliptical arena. There are 76 entrances into the amphitheatre to allow crowds to arrive and leave safely and quickly.  The architects had recognised the need for more permanent seating in the Colosseum as opposed temporary wooden benches or the piles of earth used in previous amphitheatres located in other cities. This was after all the centre of the known world and home to the Emperor and so had to be built on a grand scale never seen before. Firstly it had to be a safe place for people to visit and stay for periods of time. Many revolutionary safety devices were designed and attached to the Colosseum, such as the podiums that were built with top rollers that prevented the wild animals and convicts from climbing on top and into the crowd.  There is also the design of the exterior to consider. Despite the main activity going on inside the theatre, the design of the outside was equally if not more important. What it outwardly showed was very important to the Romans. The games were responsible for ridding the city of criminals and dangerous animals, it had to symbolise great power and splendour. The grand scale of the Colosseum was impressive but not overpowering, it was important that it was welcoming but large enough to entertain audiences in there thousands. PART TWO Essay Although they occupied one of the lowest rungs on the roman society ladder, Gladiators were widely regarded as some of the bravest members of roman society.  A gladiator was considered a professional fighter and apart from fighting other gladiators, would also pit his skills against animals in amphitheatres all over the empire. These battles started out as mere ceremonial bouts at funerals before evolving over 600 or so years into the sort of grand entertainment that we now think of as a typical day in the colosseum or indeed any of the amphitheatres around the republic and empire. The word Gladiator comes from the Latin gladiatores meaning swordsman, which in turn comes from the word gladius which was a short sword used by soldiers in the roman army. Gladiators were usually slaves or prisoners who were bought by a manager and trainer of gladiators (known as a lanista) in order to be trained as proper gladiators. There were also some free men who actually volunteered to be gladiators, no doubt looking for the celebrity that the profession offered successful entrants, a lot like children of today aspiring to becoming a famous footballer or cricketer. There were several different types of gladiators who were trained to excel in the use of different weapons, and wore different types of armour depending on the type of combat they were to enter. Some gladiators who had been prisoners of war used their native weapons and armour, and portrayed themselves as their native characters in battles, such as Gauls or Thracians.  The image often given of the Gladiator as a savage fighter might very well be just fiction. They were very skilled at what they did and like most people they would rather live than die. This is not so the case of criminals who fought and although given training, were not expected to live beyond a year. A gladiator who survived over three years was on occasion set free. It is now thought that fights may have been more civilised and theatrical than previously believed. The level of training these men undertook suggests that they would have been very good at putting on a show for the paying public and controlling movements, in the same way that the wrestlers of today are basically putting on a show. And even if the audience did order a gladiator dead at the conclusion of a match, it is highly probable that the opponent imposed only a superficial wound, in order to please the crowd. The losing gladiator might have then been dragged under the staging area and killed by an executioner undercover or allowed to recover and then resume his occupation a few months later under a new name. After all, it was all about entertaining the masses and making money for both the trainer and the owners of the venues. Because they were such expensive investments, gladiators were allowed the very best food and received the very best medical care available. In most cases Gladiators only fought 3 or 4 times a year so to stay fresh and make a big draw for the crowd who would undoubtedly had favourites from each troop of fighters who would roam from area to area fighting in different amphitheatres month after month. The games though were not only a source of entertainment for the citizens of the Roman empire, but many saw the chance to acquire fame, popularity, a reversal of lost fortunes and even freedom if they were lucky enough to survive. The games were primarily important because of what they offered those involved. Many people profited from the games, in particular those who hosted them. The games conveyed the habits of the upper class, for anyone who hosted the games it was a measure of their wealth as the expense was huge due to the number of beasts and fighters needed to entertain the crowd for any length of time. A new generation of traders and politicians found fame and popularity because they were able to spend great amounts to stage the games, which increased their status and influence within the upper social circle of the roman class system. Because of the way the games were held, it allowed the common people to mix with the upper classes at these events and on occasion win competitions for various luxury items, rather like the 1/2 time golden ticket draw held at most sporting events today. The games were not loved by everyone in roman society. As Cicero questions the measure of enjoyment the games can offer in C7 Pompeys shows, But what pleasure can it be to a man of refinement when either a powerless man is torn by a powerful beast, or else a magnificent beast is spitted on a hunting spear? What he is saying is if youve seen one man kill a beast youve seen it all and vice versa. The gladiator was looked upon as both a hero and a rogue depending on his route into the games and his ability to wow the crowd with his skills as both a warrior and an actor.  In conclusion, the gladiators were important because of what they could offer both the public and the wealthy in respect of status and prestige. The games were also important to Romans because of where they were held and what the amphitheatre conveyed, the very symbol of the politics of Rome.

Low MicroRNA-21 Expression in HPV-Induced Carcinogenesis Low microRNA-21 expression in the development of a favorable microenvironment for HPV-induced carcinogenesis Introduction Human papillomaviruses (HPVs) are the most common sexually transmitted agents. High risk types of Human Papillomavirus, as HPV16, are the causative agents of virtually all cases of cervical cancer and a significant proportion of other anogenital cancers, as well as some head and neck cancers [1-3]. The K14-HPV16 transgenic mice model is specially used to study the HPV-associated squamous cells cancers. In this model, the expression of early region genes (E2-E8) of HPV-16 is driven by the keratin 14 promoter/enhancer [4]. The K14-HPV16 transgenic mice develop epidermal hyperplastic lesions that progress to dysplastic lesions and ultimately to invasive cancer. The expression of HPV oncogenes E6 and E7 induces epithelial carcinogenesis through well-defined premalignant stages before de novo carcinoma development [5]. The basal cells are mitotically active and thus may develop further mutations in response to a proliferative stimulus, and the expression of K14 has been shown to persist in well-differentiated squamous carcinomas [6]. This model of multistep epithelial neoplasia will facilitate the study of both the epigenetic and the genetic factors that regulate neoplastic progression and coordinate malignant conversion. MicroRNAs (miRNA) are small, noncoding RNAs that regulate gene expression by base pairing with mRNAs, leading to the inhibition of mRNA translation or its degradation. In normal cells, miRNAs control numerous processes including proliferation, differentiation and apoptosis [7]. Furthermore, these molecules are described as key regulators in many diseases including, neurological disorders, cardiovascular diseases, viral infections and cancer. Some miRNAs are lost during tumorigenesis whereas other miRNAs are upregulated. Previous data indicates that miRNAs are important to distinguish subtypes of cancers, where the histological diagnosis is complex and difficult. Furthermore, it also may be a useful tool to diagnose cancers of unknown origin and to study cancer predisposition [8]. MicroRNA-21 (miR-21) has been implicated in various aspects of carcinogenesis. In most solid tumors, miR-21 is overexpressed and thus influences cell proliferation, differentiation and apoptosis [9-11]. However, the interplay between miRNAs, human papillomavirus (HPV) genes and how these interactions contribute to HPV-associated cancers remain elusive and not well understood. The tumor microenvironment associated to miRNAs plays an increasingly appreciated role in cancer (ref whiteside) however, the microenvironment prior to tumorigenesis can influence the carcinogenesis process. In this study we investigated the expression profile of microRNA-21 in K14-HPV16 transgenic mice, using ear and chest skin samples. Thus, we evaluate the importance of the microenvironment associated with the miR-21 expression and predisposition to HPV-induced carcinogenesis. Material and methods Transgenic mice K14-HPV16 mice on a FVB/N background were generously donated by Drs. Jeffrey Arbeit and Douglas Hanahan, from the University of California, through the USA National Cancer Institute Mouse Repository. Generation of K14-HPV mice has been previously reported [12]. After one week quarantine, the animals were kept as breeder pairs in accordance with National (Portaria 1005/92 dated October the 23rd) and European (EU Directive 2010/63/EU) legislation, under controlled conditions of temperature (23 ±2 ºC), light-dark cycle (12h light/12h dark) and relative humidity (50 ±10%), using hardwood bedding. A standard diet (Global Diet 2014, Harlan, Barcelona) and water were provided ad libitum. Health checks were performed daily. Genotyping of HPV-E6 and E2 (referencias genotipagem Hugo?) 15 offspring females from consecutive litters were genotyped at weaning, using tail tip samples. To genotyping were used tail tips of mice of the strain FVB, wild-type or hemizygous. Tissue lysis was performed using MAGNAPure DNA Tissue Lysis Buffer and Proteinase K for 17 h at 65  °C. Nucleic acids were extracted by the High Pure Viral Nucleic Acid Kit following the manufacturers instructions. To test the efficacy of the method of DNA extraction was investigated the presence of mouse-ÃŽ ²-globin gene. The presence of integrated HPV was assessed by amplification of HPV-E6 and HPV-E2 genes by polymerase chain reaction methodology (PCR) in-house. The resulting genotypes were confirmed to the respective phenotypes. After genotype determination, all animals were sacrificed at 22 to 26 weeks-old and completely necropsied. Ear and chest skin samples were collected into TRIzol (Invitrogen) for miRNA analysis and matched samples were collected into 10% neutral buffered formalin for histological processing. For these procedures, 13 skin samples (ear and chest) of 8 hemizygous females (+ / -) were collected. As control were used 14 skin samples of 7 wild-type females (- / -). Histology Skin samples were fixated in 10% neutral buffered formalin for 48 hours, routinely processed and paraffin-embedded. Histological sections (2 µm-thick) were obtained and stained with haematoxylin and eosin (HE) for examination on light microscopy. Samples and their lesions were classified as normal skin, epidermal hyperplasia and epidermal in situ carcinoma by two independent, blind researchers (CL and RGC). miRNA expression analysis To study miRNA-21 expression, the skin samples were macerated with the TRIzol reagent (Invitrogen) for RNA preservation. The extraction of total RNA was performed using aHigh Pure RNA Isolation Kit(Roche Applied Science), according to manufacturer’s instructions. RNA quality was assessed by measuring the absorbance at 260 nm and its purity was evaluated by the ratio of absorbance at 260/280 nm. cDNA synthesis The conversion of miRNA to cDNA was performed using TaqMan ® MicroRNA Reverse Transcription Kit ( PN 4366596 , Applied Biosystems ®, Foster CA, USA ), using sequence-specific stem-loop primers from each miRNA (miR-21 and snoRNA-202). The amplification conditions were as follows: 30 min at 15 °C, 52 min at 42 ºC and finally 10 min at 85 °C. miRNA-21 relative quantification We used qPCR technique to measure the relative expression of miR-21 (StepOne Real-time PCR Systems; final volume: 20 uL, with1Ãâ€" TaqMan ® Universal Master Mix II Applied Biosystems, Foster City, California USA; 1x MicroRNA Assay (Applied Biosystems, Foster City, California USA) and 2 uL cDNA. As endogenous control, we used snoRNA-202. Statistical Analysis Data analysis was performed by the computer software IBM ®SPSS  ®Statistics for Windows (Version 20.0). The 2ΔΔCt method, along with Student’s t-test was used in order to evaluate any statistical differences in the normalized expression of the miR-21. To analyze the normalized relative expression (-ΔCt) of the different groups, we considered the results corresponding to 99% representation of the population ( ±2).      Results Genotyping and histological analysis We observed the presence of integrated HPV DNA (E6 and E2 ORF) in 53% of mice. All cases with HPV-E6 expression also presented HPV-E2 expression. All mice with integrated HPV DNA demonstrated, phenotypically, various degrees of persistent epidermal and squamous mucosal hyperplasia, characteristic lesions associated to HPV infection, previously described in Arbeit et al. study [12]. After histologic evaluation we observed that, in all cases with integrated HPV DNA, the ear tissues presented epidermal in situ carcinomas while the chest tissues showed epidermal hyperplasia. In wild-type mice we observed normal histology. MiRNA-21 expression profile in tissue of transgenic versus wild-type mice To investigate a possible direct influence of HPV16 on the miR-21 expression profile, we analyzed the relative expression between the all tissues (ear and chest) of transgenic mice and the tissues of the control group. We did not found statistical difference in expression levels between both groups (p=0,615). When we compare the miRNA-21 expression in ear and chest samples, singly, we also did not found statistical difference in expression levels between transgenic and wild type mice (Fig.2). MiRNA-21 expression profile in normal tissue To study the miR-21 normal expression profile in tissues, we quantified the expression of miR-21 in the ear and chest skin samples of the control group. We observed that the ear tissues have lower expression levels when compared to chest tissue (p = 0.036) (Fig.3 a)). Mir-21 expression profile in tissues of transgenic mice To compare the relation between miR-21 expression and lesion type, we analyzed the histology of ear and chest samples from transgenic mice and we quantified the expression of miR-21 in the same. We detected that lower expression levels of miR-21 are associated with cancerous lesions as in situ carcinomas (ear) compared with hyperplastic lesions (chest) (p=0,043) (Fig.3 b)). Discussion Recent studies have associated miR-21 to the pathogenesis of various diseases, including cancer. (ref) MiR-21 overexpression is observed in the majority of carcinomas and hematological malignancies. However, understanding of the potential role of miRNA-21 in previous microenvironment to the development of HPV-associated lesions remains elusive. In cervical cancer, it is accepted that HPV infection is the most important factor for transition from normal cervical epithelium to cervical pre-neoplastic intraepithelial neoplasia and subsequently to invasive cervical cancer. However, the influence of others factors including the microenvironment are poorly investigated. Microenvironment associated to miR-21 may be key factor to the predisposition of cancer. We studied that the expression of miR-21 in normal tissues could be important in development of HPV-associated tumors. Our results demonstrate that, in transgenic mice, all ear tissues presented epidermal in situ carcinomas and chest tissues showed epidermal hyperplasia. Thus, we hypothesize that these interesting facts could be related to different miR-21 profile expression of both tissues. Our results indicate that there is no statistically significant difference between the miR-21 expression in HPV-positive samples and controls, concluding therefore that the presence of HPV does not directly influence the expression of this microRNA. This result is consistent with the hypothesis that differences in the miR-21 expression existing in the normal tissue microenvironment are an important determinant of the HPV-induced carcinogenesis process. Several studies have expanded the concept that inflammation is a critical component of tumour progression. Many cancers arise from sites of infection, chronic irritation and inflammation [13]. Thomas X. Lu et al., identified an IL-12/IFN-g–dependent pathway as the most prominent upregulated pathway in the lungs of OVAchallenged miR-21-/- mice compared with wild-type littermate controls, providing substantial evidence that this is the major pathway dysregulated in the miR-21–deficient mice [14]. As IL-12 is a major cytokine that regulates Th1 versus Th2 decisions primarily by inducing T cells to produce the Th1 cytokine IFN-ÃŽ ³ [15], may be related to inflammation responsible for tumor progression.
